With the continuous development of microscopic technology, users have higher and higher requirements for image quality and detail. The importance of objective lens as the core of microscopic optical system is unquestionable. In recent years, Olympus has developed several special objective lenses to meet the special needs of users for different applications to meet the experimental requirements of researchers.
Olympus XLPlan N25x, numerical aperture (NA) 1.05, water immersion objective designed especially for multiphoton imaging. It allows researchers to get deeper and clearer multiphoton microscopy images. It can be used for tissue samples with cover sheets and can also be used for patchless observations like patch clamps. Compared to other similar objectives, the objective lens has a higher Z-axis resolution and a significantly higher brightness.
The working distance of 2 mm and the angle of 35 o injection in this objective lens bring great convenience to scientific research in neurophysiology and related applications such as patch clamp. In addition, the ultra-wide field of view of 27.5 can assist researchers in obtaining more information in each multiphoton test. At the same time, the correction ring can compensate for the problem that the refractive index coefficient does not have to be matched; using this objective lens, it has excellent axial and spatial resolution of several hundred micrometers in brain tissue samples.
This is the first time that a bio-objective that meets its requirements has been developed so rapidly for the rapidly growing field of multiphoton imaging. The unique design of this objective lens will significantly improve the efficiency of multiphoton experiments. First, it corrects the spherical aberration in the range of 680-1100 nm from the Ti:Sa near-infrared laser. In addition, the new optical coating technology enables the objective lens to achieve a transmittance of 82% in the range of 400 to 1000 nm. Therefore, in the multiphoton experiment, the intensity of the excitation light is at least five times that of the same level objective lens, and can be effectively collected. More emitted light greatly increases the sensitivity and signal-to-noise ratio of the instrument.
