Experimental principle of BCA method: Under alkaline conditions, the peptide bond structure in the protein molecule can combine with Cu2+ to form a purple complex, and at the same time reduce Cu2+ to Cu+. The BCA reagent can bind sensitively and specifically to Cu+ to form a stable colored complex. The highest absorption value at 562 nm can be measured at 540-595 nm. The color depth is proportional to the protein concentration and the standard curve. In contrast, the concentration of the protein to be tested can be calculated.
